| Cell suspension cultures of chickpea (Cicer arietinum L.) were used to prepare protoplasts and vacuoles. The vacuolar preparation revealed only slight contaminations of cytoplasmic marker enzymes. H PLC analysis of the vacuolar extract showed that the malonylglucosides of isoflavones, isoflavanones and pterocarpans are exclusively located in the vacuole. Experi ments designed to determine the subcellular localization of the isoflavone malonylglucoside: malonylesterase suggest an association of this enzyme with the vacuolar membrane. Finally, a ß-l,3-glucanase and a chitinase with basic isoelectric points were also found to be localized in the chickpea vacuoles. | | |

| Isolation and Characterization of Vacuoles from Cell Suspension Cultures of Daucus carota  | | | Abstract
| A reliable procedure for the isolation o f vacuoles from anthocyanin-containing cells o f Daucus carota cell suspension cultures has been developed. From cells o f the late linear growth phase, protoplasts were prepared and purified in a sucrose/sorbitol gradient. Vacuoles were liberated from these protoplasts by osm otic shock and purified in a M etrizamide step gradient. The vacuole-containing fractions were analysed for their anthocyanin content as a measure for the yield o f vac uoles. The purity o f vacuoles was examined by assaying various marker enzym es in both proto plasts and vacuoles. The purest vacuolar fraction had 8% o f the total activity o f glucose-6-phos-phate dehydrogenase (marker for cytosol), 8% o f cytochrom e oxidase (m itochondria) and 10% o f N A DPH -cytochrom e c reductase (ER). 55% o f the acid phosphatase activity o f the protoplasts and 35% o f the total malate were recovered in the vacuoles. The vacuolar pool o f am ino acids is quite large. D ata for 15 am ino acids show that 44 to 73% are being located in the vacuole. | | |

| A Patch Clamp Study of Tonoplast Electrical Properties in Vacuoles Isolated from Chenopodium rubrum Suspension Cells  | | | Abstract
| -W ilhelm B en tru p , B ernd H offm ann , M aria G o g arten -B o ek els, Jo h a n n P ete r G ogarten The patch clamp technique has been applied to isolated vacuoles from green suspension cells of Chenopodium rubrum to record electrical parameters of the tonoplast. In a symmetrical K* solution of 46 mM, the membrane displays a near zero voltage, whereas 2 mM ATP will hyper-polarize it to 15 or 20 mV (vacuole positive). The conductance amounts to about one S-m~2. Fluctuations of the clamp current are explained by an unknown channel species having opening times of 5 — 10 ms. Together with previous work on a tonoplast vesicle preparation and unpub lished data on vacuoles from our laboratory, the present results indicate an electrogenic mem brane ATPase pumping protons from the cytoplasm to the vacuole. | | |

| Uptake of Alkaloids by Latex Vesicles and Isolated Mesophyll Vacuoles of Chelidonium ntajus (Papaveraceae)  | | | Abstract
| Latex vacuoles o f Chelidonium majus (Papaveraceae) naturally sequester protoberberine and benzophenanthridine alkaloids. Isolated vesicles take up a number o f other heterologous alkaloids, such as 9,10-dihydroergocryptine, vinblastine, strychnine, nicotine, colchicine, lupanine, and 13-hydroxylupanine, with a strong preference for the indole alkaloids and for nicotine. Kinetic data clearly show that the uptake proceeds via simple diffusion and not by carrier-mediated transport. The alkaloids are concentrated in the vesicles against a concentra tion gradient. M gA TP has no influence on alkaloid uptake. It is likely that alkaloids are trapped in the vesicles by chelidonic acid (concentration in latex 58 mM, in vesicles 661 mM) which com plexes these alkaloids and thus prevents their rediffusion out o f the vesicles. Uptake o f these alkaloids by leaf protoplasts and vacuoles o f Ch. majus, by vacuoles o f suspension-cultured cells o f Lupinuspolyphyllus and by rabbit erythrocytes was studied for comparison. | | |

| The Vacuolar Localization of Grapevine Peroxidase Isoenzymes Capable of Oxidizing 4-Hydroxystilbenes  | | | Abstract
| A study o f the subcellular localization o f peroxidase isoenzymes capable o f oxidizing 4-hydroxystilbenes to viniferin-type com pounds has been carried out in cultured cells derived from Vitis vinifera cv. G am ay berries. The study revealed that these isoenzym es are apparently located, in soluble form, in the vacuolar sap and are strongly inhibited by vacuolar anthocya-ni(di)ns in the concentration range found in planta. These results suggest that the inhibition o f the 4-H S peroxidase system in vacuoles, as a consequence o f anthocyani(di)n accum ulation, could be responsible for the loss o f viniferin production potential which accompanies grape berry veraison. | | |
