 Title    Der elektronenmikroskopische Nachweis der fibrillären Natur des Silberliniensystems bei peritrichen Ciliaten Electronmicroscopical D emonstration of the F ibrillar Nature of the Silverline System in Peritrich Ciliates  
 Abstract    A comparative light and electronmicroscopical study about the structure of the cortex, and the location and nature of the silverlines of Telotrochidium elongatum was performed. There is a fibril in the epiplasm below each pellicular ridge. Ultrathin sections of silver stained specimens demon­ strate that silver is deposited at and in these fibrills, not only in animals stained with the wet silver method but also in animals stained with the dry one. These findings confirm Klein's and our own view about the fibrillar nature of the silverline system. The significance of our findings for the elucidation of the structure and function of

 Title    Ultrastructural Changes in in vitro Ageing Spinach Chloroplasts  
 Abstract    Ultrastructural changes in in vitro ageing spinach chloroplasts have been studied in detail. Prolonged storage caused swelling of the chloroplasts due to the increase in the thickness and spacing of the thylakoid membranes. The increase in the thickness of the membrane is partly ac­ companied by the release of lipids. Addition of crystalline bovine serum albumin was found to stabilize the membrane structures. Storage of the chloroplasts at 77 °K even though it resulted in complete breakage of the whole chloroplasts, maintained the thylakoid structures in a highly intact form.

 Title    Chromoplasts in Rosa rugosa: Development and Chemical Characterization of Tubular Elements  
 Abstract    1. Development and fine structure of the chromoplasts in hips of Rosa rugosa have been in­ vestigated by electron and polarizing microscopy. 2. The chromoplasts develop from chloroplasts. During disorganization of the thylakoid system characteristic strutures become visible: 'oblique' grana, U-shaped thylakoids, and occassionally 'thylakoid teeth' as well as thylakoid anastomoses. — In this early stage, tubules appear in the plastid matrix. They are sometimes connected with membranes, especially with thylakoids. 3. In ripe hips the chromoplasts are packed with tubules. These chromoplast tubules, which attain lengths of up to at least 1.5 /um, are non-ramified and lie parallel to each other. Their diameter is variable, with a mean of 18 nm. They are polygonal in cross-section with electron-dense walls and electron-transparent cores. Association of ellipsoidal osmiophilic globules with the tubules can often be seen. Irregularly swollen remnants of former thylakoids are found in connection with these tubules. In spindle-shaped chromoplasts, the tubules are oriented parallel to the longitudinal axis. Positive birefringence and positive dichroism are observed. 4. The chemical composition of tubule fractions has been analyzed. The tubules contain ap­ preciable amounts of carotenoids, phospho-and glycolipids, and proteins. 5. The origin and development of the tubules is discussed. It appears likely that reorganization of part of the thylakoid system and synthesis of carotenoids are involved in the formation of tubules. 6. The plastids in the yellow autumn leaves of R osa rugosa contain globules but not tubules.

 Title    Ultrastructure of Differently Pigmented Synechococcus Cells  
 Abstract    Synechococcus (.Anacystis nidulans, strain L 1402-1) were grown at + 37 °C in an atmosphere o f 0.04 vol.% C 0 2 using different light conditions. Changing the culture conditions caused alterations in pigment ratios and ultrastructure o f Synechococcus. In comparison to the low white and red light grown cells under strong white light the number o f thylakoids decreased and an accumulation of storage carbohydrates could be observed. The number o f the polyhedral bodies also varied with culture conditions. The results are discussed with reference to the pigment composition and the function of the polyhedral bodies.

 Abstract    Ultrastructural features of the ciliate Hypocoma acine­ tarum, an ectoparasite of the suctorian Ephelota, led to the conclusion that the Hypocomidae do not belong to the Cyrtophorida, as some authors suggested, but should be reunified with the Ancistrocomidae in the taxon Rhyncho-dida (Chatton et Lwoff).

 Abstract    Conformational changes of the thylakoid arrangement during light-dependent etioplast-chloroplast development in cotyledons of N icotiana clevelan dii X N. glu tinosa are cor­ related with a decrease of the iron and phosphorus content in electron-dense stroma inclusions. Parallel to the trans­ formation of the prolamellar body and the stacking process of the thylakoids, both the iron and phosphorus content of the inclusions were found to be reduced. Their elemental composition was analysed by means of the energy-dispersive X-ray microanalysis. Due to their high electron-density these stroma inclusions can be observed by conventional transmis­ sion electron microscopy in unstained thin-sections from ex­ clusively glutaraldehyde-fixed material. They seem to be in­ volved in membrane formation processes concomitant with the dispersal of the prolamellar bodies. Thus, the iron and phosphorus containing inclusions were found either closely surrounded by membranes or in the intralamellar space of plastids from plantlets illuminated for 1 —8 hours. In chloro-plasts (illumination period 12 —24 hours) no connections between these inclusions and the thylakoids were noticed.

 Title    The Anatomy of the Sarcoplasmic Reticulum in Vertebrate Skeletal Muscle: Its Implications for Excitation Contraction Coupling  
 Abstract    The sarcoplasmic reticulum in situ is an intricate tubular network that surrounds the con­ tractile material in striated muscle cells. Its topographical relationship to other intracellular com ­ ponents, especially the m yofibrils, is rather rigidly m ainiained by a cytoskeleton w hich enm eshes Z line material and sarcoplasmic reticulum and, ultim ately, is anchored at the plasm alem m a. As a result, the two main components o f the sarcoplasm ic reticulum, the junctional SR and the free SR, retain their typical location in the A band region and in the I band region, respectively. The junc­ tional SR, which is thought to be the site for calcium storage and release for contraction, is, thus, always well within one micron o f the regulatory proteins associated with the actin filaments. The junctional SR, a synonym for terminal cisterna applying to both skeletal and cardiac muscle, is generally held to be involved in the translation o f the action potential into calcium release, m ainly because o f the close topographic apposition between the junctional SR and the plasm alem m a, es­ pecially in skeletal muscle. This attractive structure-function correlation is challenged by the o b ­ servation that in bird cardiac muscle 80% o f the junctional SR is spacially far rem oved from plas­ malemma, the site o f electrical activity. This anom alous topography is not in conflict with the notion that translation o f the action potential into calcium release m ay be accom plished by a d if­ fusible transmitter substance, e.g. calcium. Any hypothesis dealing with this problem must ac­ count for the anatomy o f the bird heart.
