| A quantitative determ ination method o f gallic and protocatechuic acid in cultures and liquid nutrient o f Phycom yces blakesleeanus was described. Both phenolic acids were separated by TLC and the colour reaction with Folin reagent was used for a colorimetric test. This procedure was em ployed for investigating the formation o f gallic and protocatechuic acid in cultures with opti mal (10-4 m) and reduced (1.3 x 10-6 m) zinc supply showing that their production is stim ulated by zinc ions. In addition, the inhibiting effect o f light on the accum ulation o f gallic acid was m anifested, how ever, its excretion into the m edium was uneffected by light and protocatechuic acid was not ex creted at all. During the developm ent o f Phycomyces gallic and protocatechuic acid could be detected in two days old m ycelium . With the sporangiophore production both acids are accum ulated more rapid ly in the sporangiophores. After the end o f sporangiophore formation the gallic acid content in creases only slightly. In contrast the total content o f protocatechuic acid decreases sharply. As no excretion occurs a degradation o f at least protocatechuic acid must be taken into consideration. | | |

| Inhibition of Carotene Biosynthesis in Cell Extracts of Phycomyces blakesleeanus  | | | Abstract
| Cell extracts o f the C l 15 (^-carotene-accum ulating) strain o f Phycom yces blakesleeanus were incubated with either [2-l4C]MVA or [1-I4C]IPP and a range o f possible inhibitors o f caro-tenogenesis, including bleaching herbicides, biphenyl com pounds and geranylacetone. Several o f these compounds were potent inhibitors o f /7-carotene form ation and caused the accum ulation o f phytoene. N o other carotenes were found to accum ulate, in vitro. The structures o f these inhibi tors, compared to that o f phytoene, suggest that they affect the enzym ic activity o f "phytoene dehydrogenase", possibly by com petitive inhibition. | | |
