 Title    Phospholipids and Glycerides Composition during Spheroplasts Formation of Mycobacterium smegmatis ATCC 14468  
 Abstract    The phospholipids and glycerides composition of spheroplasts of Mycobacterium smegmatis ATCC 14468 was examined. The percent total phospholipids in total lipids as well as cardio-lipin were found to be higher in spheroplasts as compared to their parent forms. Increase in cardiolipin and free fatty acids content and decrease in triglycerides levels were observed during spheroplasts formation. The results suggest that increase in cardiolipin content in sphero­ plasts is an adaptational change concomittant with the loss of cell walls.

 Title    Spontaneous Formation of Giant Phospholipid Vesicles  
 Abstract    The spontaneous formation of giant (diameter > 10 pm) vesicles from a number of phospholi-pids in excess aqueous solution has been studied by light-microscopy. Electrically neutral as well as charged phospholipids swell to form giant vesicles only if the lipids are in the fluid phase. This shows that electrostatic repulsion alone cannot explain the spontaneous formation of giant vesi-cles. The results confirm the suggestion that steric forces between extended membranes play a significant part in this process.

 Title    Phospholipids in Mediterranean Cephalopods  
 Abstract    Polar lipids of the cephalopods Eledone moschata, Sepia officinalis and Todarodes sagittatus mantle, represent 50.5%, 66.1% and 74.2% of wet tissue respectively. On the other hand the polar lipids of these three species of cephalopods constitute o f 80.8%, 94.8% and 93.7% of phospholipids, respectively. The main phospholipids identified were phosphatidylcholine (52.2, 51.3 and 58.4% of total phospholipids respectively in the above m entioned species), phosphatidylethanolamine (18.1, 19.7 and 23.9%), sphingomyelin (10.7, 15.2 and 6.7%), lyso-phosphatidylcholine (3.1, 3.8 and 1.8%) and the unusual lipid ceramide am inoethylphospho-nic acid (15.9, 10 and 9.2%). The 56.8% of phosphatidylcholine in E ledone m oschata, the 46% in Sepia officinalis and the 74.1% in Todarodes sagittatus refer to the structure of 1,2-diacyl-glycerocholine and the remaining percentage refer to the structure of l-oalkyl-2-acyl-glycerocholine or l-o-alk yl-l-enyl-2-acyl-glycerocholine. The 87.2% of phosphatidylethanolamine in Eledone m oschata, the 81% in Sepia officinalis and the 90.7% in Todarodes sagittatus refer to the structure o f 1,2-diacyl-glyceroethanolamine and the remaining percentage refer to the structure of l-o-alkyl-2-acyl-glyceroethanolam ine or l-o a lk y l-l-enyl-2-acyl-glyceroethanolamine. The major saturated fatty acids in phosphatidylcholine and phosphatidylethanolamine were C16:0 (30.3-67.5% and 23.2-54.5%) and C18:0 (3.6 -1 7 % and 1 5.4-28%), respectively, while the major unsaturated fatty acids in these lipids were C18:ln-9, n-7 (1.0-7.3% and 5.3-10.5%), C20:5n-3 (1.5-9.8% and 4 ,5-15.8%) and C22:6n-3 (12.5-42.0% and 7 .0 -11.3%), respectively.

 Title    On the Phospholipase Activity in Bovine Seminal Vesicles and Its Possible Role in the Regulation of the Prostaglandin Biosynthesis  
 Abstract    A phospholipid mixture was isolated from bovine seminal vesicle tissue. This phospholipid pre­ paration contained the following prostaglandin precursor fatty acids: 4.7 percent 5,8,11-eicosa-trienoic acid, 9.1 percent 5,8,11,14-eicosatetraenoic acid and 0.9 percent 5,8,11,14,17-eicosa-pentaenoic acid. With this phospholipid preparation as substrate phospholipase activity could be detected in different fractions of bovine seminal vesicle cells. The highest phospholipase activity was found in the microsomal fraction, less in the mitochondria and essentially no activity in the cytosol. With a microsome preparation (acetone powder) of bovine seminal vesicles the influence of certain effectors on the phospholipase activity was investigated. Prostaglandins E2 and F2a (10 5 m) showed 19 and 41 percent inhibition, DB-c-AMP (2 x 10—9 m) 54 percent, c-AMP (1 0 ~ 9 m) 22 percent, epinephrine (10—4 m) 60 percent, testosterone (10—5 m) 25 percent inhibition of the phos­ pholipase activity. Whereas ethanol exhibits also a strong inhibition on the phospholipase activity, it shows a pronounced activation of the prostaglandin synthetase.

 Title    Sinapine as a Supply of Choline for the Biosynthesis of Phosphatidylcholine in Raphanus sativus Seedlings  
 Abstract    Biosynthesis o f phosphatidylcholine in young seedlings o f Raphanus sativus is supplied with choline from degradation of the seed constituent sinapine (sinapoylcholine). This conclusion has been deduced from the following results: (1) Raphanus sativus seedlings accumulate approx. 70 nmol phosphatidylcholine which may be relevant for the consumption of choline, liberated during hydrolysis of approx. 130 nmol sinapine. (2) [14C]choline and [14C]ethanolamine, taken up by germinating Raphanus seeds, are exclu­ sively consumed in the biosynthesis o f phosphatidylcholine and phosphatidylethanolamine, respectively. (3) Pulse-chase experiments with seedlings at different germination stages show a reduced [14C]choline incorporation into phosphatidylcholine at the time when degradation o f sinapine occurs, obviously as a result of an isotope dilution by an increase o f the endogenous choline pool. (4) After [14C]choline pulse to immature seeds, during the process o f seed maturation most of the activity taken up is incorporated into accumulating sinapine and approx. 50% compared to this into phosphatidylcholine. During seedling development the quantity o f labelled sinapine rapidly decreases as a result of sinapine degradation with a concomitant label increase in free choline, phosphorylcholine, and phosphatidylcholine. Approx. 50% of the choline liberated from sinapine, is consumed in the biosynthesis of phosphatidylcholine. High performance liquid chromatographic analyses of phosphatidylcholine during Raphanus germination revealed that this phospholipid might be a metabolically active compound. Changes in the absorptivity of this compound at 210 nm indicate changes in the degree of unsaturation in the fatty acyl groups.

 Title    X-Ray Studies on Phospholipid Bilayers. II. Polymorphic Forms of Dipalmitoyl Phosphatidylethanolamine  
 Abstract    Oriented films and powder samples of the phospholipid L-x-dipalmitoyl phosphatidylethanol­ amine (DPPE) were studied by X-ray fiber diffraction techniques. The specimens were photographed under a complete range of hydration at room temperature. Two polymorphic forms were found. One of them was present in oriented films. It showed a close packing of DPPE molecules lying parallel to the normal to the bilayer plane. The other form, observed in the powder samples, had the molecules tilted by about 25°. Both forms were characterized by their unit cell dimensions, space groups, molecular conformations and packing arrangements.

 Title    Jojoba (Simmondsia chinensis [Link], Schneider)  
 Abstract    Phospholipids from Jojoba oil were isolated in amounts of 0.16%. The following phospholipids were identified: phosphatidylcholine 45%, phosphatidylethanolamine 38%, phosphatidylinositol 10% and phosphatidylglycerol 7%. The fatty acid composition is similar in all individual phos­ pholipids. Palm itic acid and oleic acid are the dominating fatty acids.

 Title    How Different are the Crystal Structures of Chiral and Racemic Diacylphosphatidylethanolamines?  
 Abstract    Both chiral and racemic phosphatidylethanolamines are known to crystallize in a similar poly-morphic form with nearly the same lamellar spacing; yet published lamellar X-ray diffraction intensity data for those materials do not agree with one another, even though the peak positions in Patterson maps are nearly the same. Translational structural searches based on the crystal struc-ture of the racemic compound also lead to similar packing models with both data sets, although the agreement between model and observed data is poor for the chiral compound. A separate analysis of L-DMPE based on lamellar electron diffraction data again leads to a similar lamellar structure with a better agreement between calculated and observed structure factors. The major difference seen for enantiomeric vs. racemic compounds is that, for the racemic lipid, the lateral unit cell spacings are about 3% larger than the chiral form, perhaps indicating a more stable hydrogen bonding network, in agreement with the higher melting point of the racemic compound. Attempts to explain this difference with other head group conformations, however, have not yet produced an improved structural model.

 Title    X-Ray Studies on Phospholipid Bilayers. VIII. Interactions with Chlorpromazine * HCl  
 Abstract    Chlorpromazine is a widely used phenothiazine tranquilizer known to alter the shape of normal erythrocytes and their osmotic fragility. In order to understand the nature of the interactions chlorpromazine • HCl (CPZ HCl) was made to interact with phospholipid bilayers formed by dimyristoylphosphatidylethanolamine (DMPE) and dimyristoylphosphatidylcholine (DMPC). This study was carried out by X-ray diffraction on crystalline powders of various molar mixtures of CPZ -HCl with DMPE and DMPC, with and without water. It was found that CPZ -HCl significa-tively affects the bilayer structure of DMPC in the presence of water, but not that of DMPE.

 Title    Determination of Glycolipids, Sulfolipid and Phospholipids in the Thylakoid Membrane  
 Abstract    D eterm ination of binding o f antibodies to lipids onto the surface o f the thylakoid mem­ brane, before and after the removal of the CF,-complex with sodium bromide, showed that in the immediate vicinity of CF, sulfolipid and monogalactolipid occur in higher concentration and are therefore arranged in domains. The m olar ratio of the CF,-complex to glycolipids was determined in Nicotiana tabacum chloroplasts o f different structure. Thus, in the chlorophyll-deficient tobacco m utants N. tabacum Su/su and Su/su var. Aurea, the m olar ratio o f C F ,/ m onogalactolipid is the same and found to be 1:570. The structure o f the lamellar system in these m utants is characterized by a higher ratio o f strom a lamellae to grana stacks when com ­ pared to the green wild type. In the wild type the ratio CF,/m onogalactolipid is 30 per cent larger (1 :740). In contrast to this the m olar ratio CF,/sulfolipid and CF,/digalactolipid is the same in the wild type and the Su/su mutant, whereas these ratios are twice as high in the yellow m utant Nicotiana tabacum Su/su var. Aurea. The binding of glycolipids and phospholipids onto the subunits o f CF, from Spinacia olera-cea was determined in the Western blot procedure by using monospecific antisera. These ex­ periments lead to the result that the two large subunits (a and ß) are m arked by antisera to monogalactosyldiglyceride, digalactosyldiglyceride and sulfoquinovosyldiglyceride. The anti­ sera to phospholipids react differently: whereas the antiserum to phosphatidylinositol only reacts with the a-subunit, the antiserum to phosphatidylcholine and that to phosphatidylgly-cerol react just as the antisera to glycolipids with both large subunits. It is observed that the antiserum to monogalactolipid occasionally m arks the y-subunit. This might mean that the glycolipids and the respective phospholipids are tightly bound onto the reacting a-and ß-subunits of the CF,-complex. Incubation of the subunit CF, with lipase from Rhizopus arrhi-zus and with phospholipase C from Chlostridium perfringens after their transfer to the nitrocel­ lulose membrane abolishes the positive reaction of the peptides with the antisera to glycolipids and phospholipids.

 Title    Quantitative Changes of the Lipid and Fatty Acid Composition of Leaves of Aleurites montana as a Consequence of Growth under 700 ppm C 0 2 in the Atmosphere  
 Abstract    Leaf lipids of Aleurites plants that were cultivated for 5 months in air containing 700 ppm C 0 2, were compared to those of control plants cultivated at 350 ppm C 0 2. The content of ether soluble lipids referred to dry matter is the same in C 0 2-and control plants. The com ­ parison of lipids analyzed as the pigments chlorophyll and carotenoids, phospholipids and glycolipids shows that the ratio of phospholipids and glycolipids is slightly shifted in favor of phospholipids in C 0 2-plants. Thus, within the group o f phospholipids, phosphatidylglycerol and phosphatidylinositol occur in higher concentrations in CÖ2-plants. Although the differences in the lipid content appear moderate in C 0 2-and control plants, it is the saturation degree of fatty acids that differs substantially. The fatty acids of C 0 2-plants contain according to the higher phospholipid content approx. 5% more saturated fatty acids. Stearic acid is three-fold increased. W hereas in the phospholipid fraction saturated fatty acids comprise one half of all fatty acids, the unsaturated fatty acids make up for 80 to 90% in the glycolipid fraction. In C 0 2-plants not only in the phospholipid fraction but also in the glycolipid fraction saturated fatty acids occur in a higher portion. This means that not only in the cell membrane of C 0 2-plants but also in the thylakoid membrane the fluidity is decreased. Also in the wax-fraction long-chained carbonic acids with 2 0 -2 6 carbon atoms occur. A s the portion of these carbonic acids is twice as high in C 0 2-plants, it is concluded that a stronger formation of the wax layers exists in C 0 2-plants. By means of Western blotting and by the use of lipid and carotenoid antisera the binding of lipids onto proteins of photosystem II and photosystem I was analyzed. It is seen that besides the major amount of lipids which build up the thylakoid membrane, som e lipids are also bound to membrane peptides. Whereas monogalactolipid is bound to the LHCP-com-plex peptides, to the O E C r peptide and the 43 and 47 kDa chlorophyll binding peptides, the anionic lipids sulfoquinovosyldiglyceride and phosphatidylglycerol and digalactolipid are bound to the core peptides of PS II and PS I. ß-carotene and the xanthophylls were found to be bound to the core peptides and ß-carotene and violaxanthin were also bound to the light-harvesting pigment complex.  Title    A Quantitative Evaluation of the Lipid Composition of Leaves of Aleurites montana as a Consequence of Growth under 0.3 ppm S 0 2 in the Atmosphere  
 Abstract    Gassing during 14 days of Aleurites plants with 0.3 ppm S 0 2 leads to quantitative modifica­ tions of the lipid composition of leaves. The ratio of phospholipids to glycolipids is shifted from 1:3 in control plants to 1:1.8 in S 0 2-plants. Glycolipids decrease from 62% of total lipids in control plants to 50% in S 0 2-plants. On the other hand phospholipids increase from 20% in control plants to 28% of total lipids in S 0 2-plants. This implies that not only the thylakoid

 Title    Phosphonolipids in the Mussel Mytilus galloprovincialis  
 Abstract    One of the possible roles of phosphonolipids is that they have a contribution to the protec­ tion of cellular integrity and survival of aquatic organisms (mollusca, cnidaria) as these lipids are included at high percentages. The total lipids of the edible mussel Mytilus galloprovin­ cialis (Mollusca, Bivalvia, M ytilidae) were found to constitute 1.27% of fresh tissue. Polar lipid com ponents constitute 61.5% of the total lipids. After separation by Solid Phase Extrac­ tion, the polar lipid fraction was separated by two dimensional thin-layer chromatography and the total phosphorus of each component, was determined. The main polar lipids found were: Phosphatidylcholine, 41.6 ± 0.8% (of which 11.3 ± 0.5% was glyceryl ether analog); ceramide am inoethylphosphonate 11.2 ± 0.2% plus 2.8 ± 0.1% another minor species; phos-phatidylethanolamine, 26.6 ± 0.5% (of which 12.2 ± 0.3% was glyceryl ether analog). The individual ceramide aminoethylphosphonate species were isolated by preparative thin layer chromatography and the structure of the major one was confirmed by a combination of analytical and chromatographic methods. Saturated fatty acyl groups with 16 carbon atoms were the main com ponents (48.4%) of the major ceramide aminoethylphosphonate species. Diglyceride aminoethyl phosphonates were not found in lipids of M. galloprovincialis.

 Title    Effects of Phospholipids in the Action of Acetyl-CoA Carboxylase from Rat Liver  
 Abstract    Acetyl-CoA carboxylase (E.C. 6.4.1.2) was isolated from rat liver. The purified enzyme con­ tains phospholipids with a rather large amount of phosphatidylinositol (26%). Incubation of the purified acetyl-CoA carboxylase with phospholipase A2 (E.C. 3.1.1.4) or with phospholipase D (E.C. 3.1.1.4) diminishes the phospholipid content by 70%, this treatment leading to a complete inactivation of the enzyme. After removal of the phospholipases, the lipid-depleted enzyme can be reactivated to a certain degree by incubation with a phospholipid extract from rat liver, with phosphatidylinositol alone, or with serum albumin.

 Abstract    A structural study o f the synthetic phospholipid, L-a-dimyristoyl lecithin (DM L), has been m ade by X-ray fiber diffraction methods. Three different types o f oriented specimens were prepared. They were X-ray photographed under the same conditions, including tem perature and relative humidity. T hree different types o f diffraction patterns, corresponding to different confor­ mations and/or packing arrangements, were found. They are characterized by their unit cell dimensions, space groups, molecular conformations, and packing arrangements.

 Title    Analysis of the Seed Lipids of Aleurites montana  
 Abstract    The seed lipids of the Chinese tung-oil tree Aleurites montana are composed of 97.2% triglycerides, 2% phospholipids and of 0.8% glycolipids. In the triglycerides 67% of all fatty acids are a-eleostearic acid (configuration C 18:3, A9 cis, A11 trans, A 13 trans). This acid is bound in the triglycerides in the 1,3 position. Via mercury Il-acetate adducts and subsequent chromatography on silica gel a separation of this fatty acid from the seed fatty acids is possible. By this procedure an a-eleostearic acid-free tung-oil mixture is obtained, which consists by one half of C i8:2, 25% of C 18:1 and by 25% of the saturated fatty acids C 16;0, C 18:0 and

 Title    Specific Positional Distribution of Acyl Moieties in Phospholipids is not Generally Deleted in Neoplastic Cells  
 Abstract    The distribution o f acyl moieties at sn-1 and sn-2 positions of cholinephosphoglycerides (CPG) and ethanolaminephosphoglycerides (EPG) has been determined for neurosarcoma, sarcoma 180 and leukemia L 1210. In all the three samples, the positional distribution o f acyl moieties in the two major classes o f phospholipids is found to be similar to that in cellular phospholipids o f most mammalian tissues. The saturated acyl moieties are located predominantly at sn-1 and polyunsaturated acyl moieties at sn-2, whereas the monounsaturated acyl moieties are randomly distributed between these two positions. Apparently, a disruption of specific positioning o f acyl moieties in phospholipids, which hitherto has been in neoplasia, does not exist in all neoplastic cells.
