| The effect o f irradiation on the anti Ig binding ability o f lymphocytes after irradiation was studied. Norm al control and irradiated lym phoid cells were treated with rabbit anti mouse IgG fluorescein conjugated serum and the fluores cein positive cells compared. A reduction in the proportion o f stained cells in the irradiated lym phocytes was found. It depends on the dose and on the incubation tim e at 37 °C after irradiation. The m axim um effect was detected after 10 minutes o f incubation, longer incubation is associated with a partial recovery o f the stainability o f the IgG recep tors. | | |

| Induction of DNA-Protein Cross-Links by Platinum Compounds  | | | Abstract
| The differences between cis-and mws-diamminedichloroplatinum II (DDP) in forming DNA-protein cross-links in isolated human lymphocytes were investigated. Both cis-and rra/75-DDP can induce DNA-protein cross-links. We show that cis-DDP forms complexes between D N A and proteins faster than trans-DDP. This results from an increase in the quan tity of D NA and platinum together with an increase in drug concentration. Under the same conditions trans-DDP causes a decrease in DNA-forming complexes with proteins. After a 12 h incubation of lymphocytes we observe a similar level of DNA in DNA-protein cross links induced by D D P isomers, but more platinum appears in complexes induced by trans-DDP. The results obtained demonstrate that the antitumor drug -ds-DDP and the clinically ineffective trans-DDP induce links between D N A and proteins in a different manner. We suggest that the therapeutic activity of ds-DDP can in part arise from rapidly forming DNA-protein complexes which can destroy the most important cellular processes, such as replica tion and transcription. | | |

| During persistent infection of mice with the lymphocytic choriomeningitis (LCM) virus approximately 3% of leuko cytes were found to contain viral antigen and to produce infectious virus. Morphologically, infected cells were shown not to be lymphoblasts and their numbers were not reduced by removal of mononuclear phagocytes. We conclude that in LCM virus carrier mice true lymphocytes participate in the infectious process. | | |

| The Influence of Spermine, Spermidine and Various Sera on T-Lymphocyte and Granulocyte Colony Growth in vitro  | | | Abstract
| Using T-lymphocyte (T-LC) and granulocyte colony (GC) assays with truly proliferating cells, the dose-response relationships of spermine and spermidine, of fetal calf (FCS), calf (CS), horse (HS) and human AB serum (A B S), and of the polyamines in the presence of selected does of the sera were analyzed. In contrast to earlier observations using [3H] thymidine uptake measure ments it was found that the polyamines as well as the sera themselves inhibit T-LC and GC growth in the presence of autologous HS and ABS, respectively. The polyamines also inhibit in the presence of additional HS and ABS, yet most effectively with added FCS and CS. Thus the previously reported species-specificity of the serum-dependent polyamine inhibition has here been shown to be more quantitative than qualitative. These studies stress the significance of assays utilizing truly dividing cells and broad dose-response relationships in order to assess specific biological effects in vitro. | | |
