 Title    Isoflavone and Pterocarpan Malonylglucosides and ß -l ,3 -G l u c a n -and Chitin-Hydrolases are Vacuolar Constituents in Chickpea (Cicer arietinum L.)  
 Abstract    Cell suspension cultures of chickpea (Cicer arietinum L.) were used to prepare protoplasts and vacuoles. The vacuolar preparation revealed only slight contaminations of cytoplasmic marker enzymes. H PLC analysis of the vacuolar extract showed that the malonylglucosides of isoflavones, isoflavanones and pterocarpans are exclusively located in the vacuole. Experi­ ments designed to determine the subcellular localization of the isoflavone malonylglucoside: malonylesterase suggest an association of this enzyme with the vacuolar membrane. Finally, a ß-l,3-glucanase and a chitinase with basic isoelectric points were also found to be localized in the chickpea vacuoles.

 Title    Phenolics from Cell Suspension Cultures of Penstemon serrulatus; Relation to Plant Organs  
 Abstract    By repeated selection of pigment portions of tissue the red callus induced from root seed­ lings of Penstemon serrulatus Menz. was chosen for suspension culture, which was maintained in Schenk and H ildebrandt medium supplemented with naphthaleneacetic acid (0.2 mg/1), 6-benzylaminopurine (2 mg/1) and sucrose (50 g/1). From the cultured cells eight phenolic compounds were isolated. They were identified as cyanidin 3-O-glucoside, delphinidin 3-0-glucoside, luteolin, luteolin 7-O-glucoside, norartocarpetin 7-O-glucoside, verbascoside, mar-tynoside and leucosceptoside A. The kind of cell line, its age and light irradiation were im portant factors in flavonoid production, but production of phenylpropanoid glycosides was found to be unaffected by these factors. The phenolic composition found in the cell culture was com pared with those in the flowers and leaves

 Title    Influence of Medium Composition on the Formation of Secondary Compounds in Cell Suspension Cultures of Catharanthus roseus (L.) G. Don  
 Abstract    Cell suspension cultures o f Catharanthus roseus have been subjected to various m edia condi­ tions in order to stim ulate the formation o f indole alkaloids. H igh ajmalicine contents (up to 0.5 m g/g cell fresh weight) were achieved by transferring 2-week-old cell suspensions to a 10-fold volum e o f a 8% sucrose solution. The alkaloid accum ulation started two days after the transfer and reached a plateau after ten days. Furthermore an enhanced level o f phenolic com pounds was found, whereas growth o f the culture was low. The accumulation o f both, alkaloids and poly­ phenols was stimulated by high concentrations o f sucrose and low concentrations o f nitrogen con­ taining salts and phosphate. W hen these m inerals were added to the sucrose solution in con­ centrations com m only used for cell culture m edia, the accum ulation o f alkaloids and phenolic compounds was largely suppressed.

 Title    Elicitor-Stimulated Furanocoumarin Biosynthesis in Cultured Parsley Cells: S-Adenosyl-L-Methionine:BergaptoI and S-Adenosyl-L-Methionine: Xanthotoxol O-Methyltransferases  
 Abstract    Cultured parsley cells (Petroselinum crispum) treated with an elicitor from the soybean patho­ gen, P hytophthora m egasperm a f. sp. glycinea, were used as source for the separation, extensive purification and characterization of S-adenosyl-L-methionine: xanthotoxol and S-adenosyl-L-m ethionine: bergaptol O-methyltransferases. The products, xanthotoxin and bcrgapten, were among the most abundant coumarin derivatives accumulated in the culture fluid of elicitor-treated parsley cells. The latter enzyme also catalyzed the methylation of 5-hydroxyxanthotoxin to isopimpinellin, another major coumarin derivative in this system. The activities of both enzymes showed transient increases upon elicitor application prior to the accumulation of the products. No activity was detectable in untreated cells. Activity increases of the O-methyltransferases occurred a few hours later than those of phenylalanine ammonia-lyase and 4-coumarate:CoA ligase.

 Title    Indole Alkaloids from Ochrosia elliptica Plant Cell Suspension Cultures  
 Abstract    From cell suspension cultures of Ochrosia elliptica Labill. eight different monoterpenoid indole alkaloids were isolated and comprised three alkaloid classes viz. Corynanthe/Heteroyohimbine (tetrahydroalstonine, cathenamine, pleiocarpamine and two methoxylated heteroyohimbines), Strychnos (norfluorocurarine), Apparicine (apparicine and epchrosine). A ll these compounds except the Strychnos alkaloid are biogenetically closely related to the known alkaloids occurring in Ochrosia plants, although such alkaloids have not previously been detected in O. elliptica. Epchrosine has not before been described as a natural product.

 Title    Major Indole Alkaloids Produced in Cell Suspension Cultures of Rhazya strict a Decaisne  
 Abstract    Eleven main alkaloids were identified from cell suspension cultures of Rhazya stricta grown in 4X-medium for 15 days. The alkaloids comprised the five groups Corynanthe, Strychnos, Ebur-nan, Secodine, and Aspidosperma and can be regarded as being typical Rhazya alkaloids, al­ though the Strychnos alkaloid akuammicine has been isolated for the first time from the genus Rhazya. The most abundant compound was (+)-l,2-dehydroaspidospermidine (15 mg/1 medium) whereas all other constituents were synthesized in about 5 — 10 times lower amounts. More than 15 further alkaloids were formed as minor components which have not yet been identified. Culti­ vated Rhazya cells have been shown to be one of the richest alkaloid sources from apocynaceous cell suspensions. The isolated compounds, because of their structural heterogeneity, cannot be presently arranged into a scheme with coherent biosynthetic sequences.

 Title    Glutaric Acid as a Catabolite of Nicotinic Acid in Parsley Cell Suspension Cultures  
 Abstract    A degradation product of nicotinic acid representing the pyridine carbon skeleton was isolated and purified from parsley cell suspension cultures after incubation with [6-14 C]nicotinic acid for 70 h. The catabolite was identified as glutaric acid by means of spectroscopic (GC-MS and 'H NMR) and chromatographic (TLC, HPLC) techniques. Glutaric acid when applied to parsley cell cultures was readily degraded to C0 2 but intermediate products could not be identified.

 Title    Comparative Investigations on the Metabolism of the Herbicide 2-(2,4-Dichlorophenoxy)-propionic Acid in Plants and Cultured Cells of Tomato

The m etabolism o f [ l -l4C ]2-(2,4-dichlorophenoxy)-propionic acid was studied in excised plants and cell suspension cultures o f tom ato. It was rapidly taken up and m etabolized by both the plants and the cultured cells. The m etabolites, isolated by extraction with aqueous acetone, separated and purified by TLC and H PLC, were identified by chem ical and spectrometric methods. C onju­ gates with carbohydrates were detected. G lucose, diglucose as the main conjugating m oiety, and triglucose were found as carbohydrate com ponents within the conjugates. Alm ost the same conjugates occurred in plants and cultured cells.

 Title    Hydroxyphenylpyruvate Reductase from Cell Suspension Cultures of Coleus blumei Benth  
 Abstract    Cell suspension cultures o f Coleus blumei Benth. producing high amounts o f rosmarinic acid were used to study the biosynthetic pathway o f this caffeic acid ester. One o f the involved en­ zymes, the hydroxyphenylpyruvate reductase (H PPR), is characterized in this paper. HPPR catalyzes the N A D (P)H dependent reduction o f /^-hydroxyphenylpyruvate to /»-hydroxyphen-yllactate. The enzyme developed maximal activity at an incubation temperature o f 37 °C and at a pH o f 6.5 to 7.0. The reaction proceeded linearly for an incubation time o f 60 min and up to a protein concentration o f 0.2 mg per assay. As electron donor HPPR accepted N A D H and N A D P H with A^m-values o f 190 jim and 95 |im respectively. The enzyme reduced differently substituted hydroxyphenylpyruvates but not ß-phenylpyruvate. The apparent A^m-values for the various substrates were at 10 for /^-hydroxyphenylpyruvate, at 130^xm for 3,4-dihy-droxyphenylpyruvate and at 250 |iM for 3-methoxy-4-hydroxyphenylpyruvate. HPPR was com petitively inhibited by rosmarinic acid and pyruvate with /^-values o f 210 |iM and 200 |IM respectively. Caffeic acid,/>-coumaric acid and cinnamic acid did not affect the enzyme activity but/>-coumaroyl-CoA inhibited HPPR.

 Title    A Model Mechanism for the Enzymatic Synthesis of Lupin Alkaloids  
 Abstract    A crude enzyme preparation obtained from cell suspension cultures o f Lupinus polyphyllus cat­ alyzes the pyruvate dependent conversion o f cadaverine into the tetracyclic lupin alkaloids. As the first reaction product 17-oxosparteine could be identified by gas-liquid chrom atography and mass spectroscopy. In som e experiments sparteine was found additionally. A participation o f di­ amine oxidase could be ruled out. The cadaverine-pyruvate transaminating enzyme system (17-oxosparteine synthase) catalyzes the formation o f 17-oxosparteine from three cadaverine units without releasing free intermediates. These results are inconsistent with the hypothetical m echa­ nism thus far formulated for the lupin alkaloid biosynthesis. A new enzym atic model mechanism is proposed regarding both the results o f the enzymatic experiments and those o f the in vivo tracer studies.

 Title    Indole Alkaloids from Cell Suspension Cultures of Stemmadenia tomentosa and Voacanga africana  
 Abstract    Cell suspension cultures o f Stemmadenia tomentosa synthesized under normal growth condition the eight major indole alkaloids: (-)-tabersonine, (—)-minovincinine, (+)-conoflorine (voaphyl-line), condylocarpine, (+)-tubotaiwine (dihydrocondylocarpine), (-)-norfluorocurarine (vin-canine), (-)-vinervine, and (—)-coronaridine. These alkaloids consist o f the three different types, Aspidosperma, Strychnos and Iboga. In contrast, cultures o f Voacanga africana produced mainly one alkaloid group (Aspidosperma-type) represented by (—)-tabersonine, lochnericine and (—)-minovincinine. Therefore this cell culture seems to be qualified for investigation concerning the biosynthesis o f Aspidosperma alkaloids.

 Title    Effects of Permeabilization on the Biotransformation of Phenylalanine by Immobilized Tobacco Cell Cultures  
 Abstract    Im m obilized tobacco cells excreted only a small proportion of their main secondary m etabolite caffeoylputrescine into the culture m edium . Experim ents designed to release more o f the com ­ pound by perm eabilization caused the loss o f caffeoylputrescine, probably by oxidative reactions. M oreover, rather mild treatments with pejm eabilizing agents (e.g. «-propanol) resulted in severe growth inhibition. The ability of perm eabilized cells to form caffeoylputrescine and other hy-droxycinnam oyl conjugates from phenylalanine decreased considerably even when such cells were still able to m etabolize phenylalanine into various ethyl acetate extractable com pounds (e.g. hydroxycinnamic acids and acetophenones). The formation of new biotransformation products suggests that perm eabilized cells could be used as a tool for testing the enzymatic capabilities o f a cell culture.
