| Complex Formation of Carnosine with Purine Nucleotides in Aqueous Solution  | | | Abstract
| This ^ -N M R study provides experimental evidence for an intermolecular interaction between the dipeptide carnosine (yS-alanyl-L-histidine) and the purine nucleoside 5'-m onophosphates 5'-AM P, 5'-IMP and 5'-GM P. From the observed upfield shifts o f the purine nucleotide and im idazole proton resonances it is concluded that the interaction is o f the stacking type and that it involves the purine base o f the nucleotide and the histidine m oiety o f carnosine. Apparent microscopic equilibrium constants and com plex shifts are obtained with a m icroscopic m odel, which considers the formation o f both 1:1 and 1 :2 complexes. The stacking pattern for com plex formation between the histidine m oiety o f carnosine and the adenine m oiety o f 5'-AM P is con structed by fitting the experimental 5'-AM P com plex for histidine. | | |

| The Specificity of Ionophore A 23187 in Cation Transport across Lipid Membranes Studies with Lecithin Vesicles  | | | Abstract
| Lecithin vesicles containing different cations were prepared by sonication and characterized. When the concentration o f the divalent cations in the buffer was increased from 1 m M to about 100 m M the cation-concentration inside o f the vesicles did not increase proportional to the outside concentration, but showed a saturation behaviour. The efflux o f various cations m ediated by ionophore A 23187 was m easured and relative transport-rates were determined. The following sequence was obtained: Zn2+ > Mn2+ > Ca2+ > M g2+ > Sr2+ > Ba2+ % L i+ > N a +. The efflux o f Ca2+ increased proportional to the square o f the A 2 3 187-concentration. To lecithin-vesicles containing ethylenediam ine tetraacetate (ED TA) inside different cations were added on the outside. In the presence o f A 23187 divalent cations are transported into the vesicles and bound there by EDTA. During the influx o f M e2+-ions into vesicles a H +-efflux was observed. The resulting pH-decrease was measured. The rate o f the pH -change depended on the M e2+-ion used. The sequence was: Cd2+ > Zn2+ > Ca2+ > M n2+ > M g2+ > Sr2+, Ba2+. | | |

The com plexes formed are relatively weak (as sociation constants 4 -4 8 M_1), shortlived (life tim e smaller than 0.02 s), and m ainly 1:1 in stoichiometry preferring the terminal phosphate o f ATP as the site o f association. The divalent metal ions M g2+ and Ca2+ bond stronger than the am ines (association constants 1500 and 100 M_1, resp.) prefer the /?-phosphate group changing thereby the geom etry o f the ATP phosphate chain. | Effects of Polynucleotides on Production and Activity of Streptolysin S  | | | Abstract
| Activity o f various natural and synthetic polynucleotides as a carrier for streptolysin S was tested in a resting cell system. As the carrier, intact m olecules o f M S2 R N A , E. coli tR N A or rat liver R N A were almost inactive, whereas R N ase I core o f these R N A s, especially the core frac tions eluted from DEA E-cellulose colum n at higher NaC l concentrations, effectively induced pro duction o f the extracellular hem olysin. The carrier activity o f yeast R N A was significantly en hanced by sim ultaneous addition o f R N ase I, to the streptococcal suspension. Preincubation o f yeast R N A with growing streptococci or a protein fraction from the bacterial culture supernatant increased its carrier activity. Evidences were obtained suggesting involvem ent o f streptococcal nuclease in the enhancement o f the carrier effect. Production o f the streptococcal hem olysin was markedly prom oted by polyguanylic acid but neither by polyadenylic acid, polycytidylic acid nor by polyuridylic acid. Like trypan blue, polyguanylic acid exerted potent inhibitory effect on hem olytic activity o f streptolysin S complex. Similar but less marked effect was observed with certain R N ase I core fractions o f rat liver R N A. Som e data concerning effect o f polynucleotides on stability o f the hem olysin were also presented. | | |

| The fluorescent staining o f fixed A 9 cells (m ouse fibroblasts) with ethidium brom ide E has been investigated qualitatively and quantitatively. E is bound to D N A , R N A and protein. After enzymatic R N A digestion and staining at pH = 4.0 the dyestuff is bound to D N A specifically. The formed E -D N A -com plex is stoichiom etric if D N A will be saturated by high dye concentra tion, CF = 1.0 x 10~2 M. The stoichiom etric factor n = 0.21 was determined from Scatchard bind ing isotherms and by spectrophotometric titration. The binding o f E to D N A and R N A has been investigated by binding isotherms. E is bound to D N A by intercalation and by ionic interaction. It is bound to R N A by ionic interaction only. The ionically bound dyestuff can be replaced by salts like LiCl from D N A and RNA. Only inter calated E remains. The com petitive salt effect can be used to avoid the enzym atic R N A digestion in the specific staining o f D N A . W ithin the cell the E-D N A -com plex fluoresces strongly. U sing a microspectrophotom eter we determined the fluorescence intensity J and the extinction E in cell sections o f q = 1 |a2 area. The fluorescence was excited in the m inimum o f the absorbance o f the E -D N A -com plex at = 365 nm. The extinction E 2 was measured in the short wavelength band at X2 = 260 nm. Under these conditions we received a linear relation J = J (E ?) up to high extinctions E 2. The quantum yield Q o f the E -D N A -com plex is nearly constant and independent o f the biological environment. Q is also unaltered by LiCl. W ithin the lim its o f error intercalated and ionic bound dye have the same quantum yield. Under the conditions o f stoichiom etric staining the am ount o f E -D N A -complex and therefore D N A in the cell can be determined by J. The extinction coefficient e2 = 45200 M-1 cm -1 o f the E -D N A -com plex at 260 nm was calcu lated from the concentration dependence o f the absorbance spectra using the law o f mass action. By e2 the am ount o f D N A in the cell is accessible from J. According to our measurements with A 9 cells the superficial density o f D N A has the order o f m agnitude £>n = m ^ /q = 101 nmol cm -2, the amount raN o f D N A per section q = 1 jj.2, mN = 10_l fmol, and the D N A concentration in the cell nucleus CN = 10-1 M. | Antigens cause an increase o f the D NA synthesis in the spleen o f mice as shown by the in corporation of [32P]orthophosphate and [3H]thym idine. In the present paper, the incorporation of [32P]orthophosphate into the single deoxymononucleotides has been studied under influence of the antigen bovine serum albumin. For this purpose, the labelled D NA was decomposed to the deoxymononucleotides and their specific activities determ ined. Using [32P] orthophosphate, it was found that the activity o f the D NA increased continuously during an observation period of 8 h. While after one hour the relative specific activity o f dTM P was highest and that o f dAMP lowest, the relative specific activities o f the deoxymononucleotides had become equal after 8 h. Under the influence of bovine serum album in, the incorporation o f [32P]orthophosphate increased in the main band DNA as well as in the satellite DNA. The antigen had no effect on the distribution of the specific activities o f the deoxymononucleotides. | | |

| The isolation of transcribed D NA sequences of P815 cells and the partial characterization with respect to their sequence composition and relative rates o f enzymatic D NA m ethylation are reported in this paper. Transcribed regions were purified by affinity chrom atography using im mobilized heterogenous nuclear RNA of P815 cells. About 10% of total genome was found in this fraction. Reassociation analyses showed differences in sequence composition o f transcribed versus non-transcribed D N A fractions. The relative proportion of inverted repeats was doubled in the transcribed fraction whereas ordinary highly repetitive sequences comprising mainly o f satellite D NA were found alm ost exclusively in the non-transcribed regions o f the P815 genome. A bout 70% of transcribed portions corresponds to unique and interm ediary D NA sequences. A fter labelling of cells with L-[Methyl-3H]methionine and [14C]deoxycytidine relative rates o f enzymatic DNA m ethylation were computed for different kinetic components o f transcribed and non-transcribed portions of P815 genome. No difference was found except in inverted repeats. In transcribed D NA the relative rate o f enzymatic DNA methylation was only about 40% o f that of the non-transcribed ones. We have quantitated this hypomethylation and found that there is, in average, about one 5-methylcytosine residue in 100 nucleotides o f transcribed inverted repeats, com pared to about 2.5 5-methylcytosines in non-transcribed fractions. In view of these data we propose that the enzymatic methylation o f inverted DNA repeats negatively controls the tran scriptional process in a given genomic region. | | |

| Chlorophyll-Protein Complexes of Chlorella fusca  | | | Abstract
| Chlorophyll-protein complexes from thylakoids of the normal type and two mutants o f Chlorella fusca were separated using sodium dodecyl sulfate acrylamide gel electrophoresis (PAGE). The properties o f the chlorophyll-protein complexes o f the three strains of Chlorella were compared. Standard curves were set up for the characterization of the chlorophyll-proteins. In every electrophoretic separation of chlorophyll-protein complexes, a certain am ount o f pigm ent is separated from the protein. We tried to keep that am ount as low as possible by mild solubiliza tion and by working in low tem perature. U nder these conditions, we obtained several new chlorophyll-proteins in addition to the P-700-chlorophyll a-protein (CP I) and the light-harvesting chlorophyll a/b-protein (CP II). Thus, a small band (CP I a) was located between the top of the gel and the CP I after elec trophoresis. A lthough it shows spectral qualities sim ilar to CP I, it possesses a much lower chloro phyll a/chlorophyll b ratio. It may be an aggregate of photosystem I and light-harvesting chloro phyll. We found and analyzed three other chlorophyll-proteins with electrophoretic m obilities inter mediate between that o f the P-700-chlorophyll a-protein and that of the light-harvesting chloro phyll a/b-protein complex. Two o f these chlorophyll-proteins, the LHCP1 and the L H C P2, have a low chlorophyll a/chlorophyll b ratio and spectral properties similar to that o f the light-harvesting chlorophyll a/b-protein (LH CP3). They obviously represent dimers or oligomers o f the latter com plex. A third, new complex (CPa) m igrated between LHCP3 and its dimer. W ith the chlorophyll b deficient m utant G 36 of Chlorella fusca, this complex could be obtained in high purity and great enrichment (15% of total chlorophyll). It could be proved that this complex only contains chloro phyll a. Its red absorption m axim um is at 671 nm. Some indirect evidences suggest that it may be a good candidate for the PS II reaction center complex. | | |

| Isolated intact spinach chloroplasts were supplied with 14C-labeled C 0 2, phosphoglycerate, phos-phoenolpyruvate, acetate and mevalonate and the incorporation o f radioactivity into /?-carotene and plastoquinone-9 assayed. All applied substrates were capable of crossing the chloroplast envelope. Except phosphoenolpyruvate all radioactive precursors were incorporated into ß-carotene and also into plastoquinone-9. It is concluded that spinach chloroplasts are autonom in acetyl-coenzyme-A-formation and terpenoid biosynthesis. There is a direct carbon flow from intermediates of the Calvin-cycle via acetyl-coenzyme-A and mevalonic acid existing in the chlo roplast leading to the carotenoids and isoprenoid side-chains o f chlorophylls and quinones. | | |

| A compound showing blue fluorescence under UV-light was isolated from spruce needles and identified as umbelliferone. Structure analysis was based on UV and mass spectroscopy as well as chromatographic comparison with authentic material. | A Series of Novel Flavanones from Fern Exudates  | | | Abstract
| The white farinose exudate on the fronds o f Cheilanthes argentea contains a series of flavanones as minor constituents. These were identified by spectroscopic methods as: 5,4'-diOH,6,7-diOMe flavanone and its 4'-OMe derivative, 5,4'-diOH,7,8-diOMe flavanone and its 4'-OMe derivative, 5,6-diOH,7,8,4'-triOMe flavanone, 5,4'-diOH, 6,7,8-triOMe flavanone and its 4'-OMe derivative. The farina o f Notholaena limitanea var. mexicana contains eriodictyol-7,4'-di-methyl ether and eriodictyol-7,3', 4'-trimethyl ether as minor components. Both compounds were also identified in the exudate of N. fendleri, together with naringenin-7-OMe and naringenin-4'-OMe. In N. lemmonii var. lemmonii trace amounts of 5-OH,7,3',4',5'-tetra-OMe flavanone were detected. Nine of these substances are novel natural flavanones. | | |

The 3-hydroxylase is a soluble enzyme and requires as cofactors 2-oxoglutarate, Fe2+ and ascorbate. Besides naringenin eriodictyol is a substrate for the 3-hydroxylase. The 3'-hydroxylase is localized in the microsomal fraction and requires NADPH as cofactor. Naringenin and dihydro-kaempferol but not 4-coumarate or 4-coumaroyl-CoA are substrates for this enzyme. 3'-Hy-droxylase activity is present only in genetic lines of M. incana with the wild-type allele b +. | | |
